NLRP3 inflammasome is a group of cytoplasmic protein complexes, which are composed of NLRP3, ASC and caspase-1. The activation of NLRP3 inflammasomes in macrophages requires two steps: priming and activation. The priming step (signal 1) is provided by inflammatory stimuli, such as TLR4 agonists, which induce NF-κB-mediated expression of NLRP3 and pro-IL-1β. The activation step (signal 2) is triggered by PAMPs and DAMPs to promote NLRP3 inflammasome assembly. The assembled NLRP3 inflammasome can activate the protease caspase-1 to induce Gasdermin D-dependent pyroptosis and promote the maturation and release of IL-1β and IL-18, which contributes to the maintenance of innate immune defense and homeostasis.

The first step (establishment of in vitro models):
Classical in vitro models of NLRP3 inflammasome activation include THP-1 and Bone marrow-derived macrophages (BMDM). We first used PMA to induce THP-1 from monocytes into suitable macrophages, or isolated bone marrow cells from mouse femurs and tibias, and induced them into BMDM cells.
The second step (induction of NLRP3 inflammasome activation):
The priming process is initiated by LPS, leading to the activation of the transcription factor NK-κB and the subsequent transcription and synthesis of pro-IL1β and NlRP3. The activation process is induced by specific NLRP3 stimuli such as Nigericin, ATP, MSU, ALUM, etc., causing the assembly and activation of NLRP3 inflammasomes, and mediating the autocatalytic activation of Caspase-1. Active caspase-1 will further cause pro-IL -1β and pro-IL-18 maturation and secretion.
