γ-secretase is an enzyme complex that plays a key role in processing several transmembrane proteins, including the amyloid precursor protein (APP) linked to Alzheimer's disease (AD). Specifically, it cleaves the C-terminal fragment C99 in the transmembrane domain of APP. This process leads to the release of various lengths of Aβ peptides, with Aβ40 and Aβ42 being the predominant forms. By inhibiting or modulating γ-secretase, it is possible to decrease the production of Aβ peptides and alleviate their harmful effects.
