Introduction

Apoptosis refers to the orderly and autonomous death of cells controlled by genes in order to maintain a stable internal environment. Apoptosis is an active process of cells, involving the activation, expression and regulation of a series of genes. It is a death process that cells actively strives to better adapt to the living environment.

The main morphological feature of apoptosis is that the chromatin is fragmented into different sizes, which gather together with certain organelles, surrounded by a reflexed cell membrane, forming apoptotic bodies. Therefore, the detection methods are different according to the different stages of apoptosis. Apoptosis is a multi-step process including early, mid and late cell events, and can be detected using various cell detection methods.

Stages of apoptosis:

Early events

  • Translocation of phosphatidylserine outside the cell membrane
  • Loss of mitochondrial membrane potential
  • Release of cytochrome C and ATP
  • Activation of caspase-8 and 9

Mid-term events

  • Activation of caspase-3, 6 and 7
  • Poly-ADP-ribose polymerase (PARP) cleavage
  • Shrinkage of cells
  • Nuclease activation

Late events

  • DNA fragmentation
  • Disintegration of the nucleus
  • Formation of apoptotic bodies
  • Phagocytosis of macrophages
