Background

Adhesion to the extracellular matrix (ECM) is essential for the survival and reproduction of many adherent cells. Apoptosis caused by loss of adhesion to ECM is called anoikis. Anoikis is involved in the physiological processes of tissue renewal and cell homeostasis.

As a special form of programmed cell death, anoikis plays an important role in the development of the body, self-balance of tissues, disease occurrence and tumor metastasis. The initiation and execution of anoikis are mediated by different pathways, all of which terminally converge into the activation of caspases and downstream molecular pathways, culminating in the activation of endonucleases, DNA fragmentation and cell death.

It is consistent with the classic apoptosis, which can be induced by the mitochondrial pathway or the cell surface death receptor pathway. Our services include but are not limited to:

MTT and Calcein AM/EthD-1 Fluorescence Detection

Calcein-AM can easily penetrate the membrane of living cells and is a cell staining reagent that can fluorescently label living cells. When it enters the cytoplasm, esterase will hydrolyze it into Calcein, which stays in the cell and emits strong green fluorescence. EthD is a red fluorescent dye that can only enter dead cells. After EthD binds to DNA, the fluorescence intensity increases by 40 times, and there is almost no fluorescence before entering the cell. Therefore, it can be used to detect anoikis cells.

Creative Bioarray provides you with a full set of cell analysis and detection solutions, and provides MTT and Calcein AM/EthD-1 fluorescence detection solutions for anoikis tests.

The principle is to spread the cells on a culture plate coated with poly-Hema (an anionic polymer that prevents cells from adhering to the wall), and measure the activity of the cells by MTT and Calcein AM. Red EthD-1 reagent is used to detect dead cells.
