AIM2 is an important inflammasome component that acts as a sensor that triggers inflammasome activation, pyroptosis, and IL-1β and IL-18 release in response to intracellularly delivered dsDNA. AIM2 senses potentially dangerous cytoplasmic DNA, and AIM2 that recognizes dsDNA recruits ASCs through PYD homotypic interactions. ASC in turn recruits pro-caspase-1 through CARD-CARD interactions, leading to the cleavage of pro-IL-1β and pro-IL-18, which ultimately leads to the secretion of mature IL-1β and IL-18.

The first step (establishing an in vitro model):
Classic in vitro models of activation of the AIM2 inflammasome include THP-1 and BMDMs. We first treated THP-1 with PMA for a period of time to induce THP-1 from monocytes to suitable macrophages, or isolated bone marrow cells from mouse femurs and tibiae and induced them to BMDM cells.
The second step (induction of AIM2 inflammasome activation):
After LPS-induced macrophages for a period of time, transfection with poly(dA:dT) for several hours fully induced the activation of AIM2 inflammasome.
Cells and supernatants were collected after stimulation. We analyze the relative protein levels of Gasdermin D, Caspase-1, AIM2, ASC, IL-1β by Western blotting. We assess LDH levels in the supernatant by an LDH assay kit. We detect IL-1β, IL-18 and TNF-α in the supernatant by ELISA. We detect relative mRNA expression of IL-1β, IL-1α, IL-18, IL-6 and TNF-α by qPCR. We examine the interaction between ASC and AIM2 by co-immunoprecipitation. We analyze the localization of AIM2 and ASC in cells by confocal microscopy.
