They have obvious characteristics of caspase activation and DNA fragmentation to a specific length, and the cell membrane remains intact during these processes. The cell membrane bulges and vesicles are formed, leading to the production of "apoptotic bodies" engulfed by macrophages or neighboring cells. Unlike necrosis, apoptosis does not trigger an inflammatory response, and neighboring cells remain intact. Double Detection (Cell Viability + Apoptosis)

We can detect cytotoxicity marked by cell viability and the apoptosis marked by caspase activation to determine cell death mechanism.

Principle:

  • Cell viability: Use cell-permeable substrates to detect live cell proteases in cells and detect fluorescent signals (400Ex/505Em).
  • Caspase-3/7 cleaves the substrate and releases amino luciferin, which emits light under the catalysis of luciferase.

Triple Detection (Cell Viability + Cytotoxicity + Apoptosis)

Creative Bioarray can simultaneously evaluate cell viability, toxicity and apoptosis events.

Detection of Cell Viability and Cytotoxicity
  • Add reagents containing two peptide substrates during the experiment.
  • The fluorescent peptide substrate (GF-AFC) (which can penetrate the cell membrane) enters the living cells and is sheared to generate a fluorescent signal. The signal intensity is proportional to the number of living cells.
  • The second fluorescent peptide base (bisAAF-R110) (cannot penetrate cell membrane) can detect the activity of dead cell protease.
Apoptosis Detection
A reagent containing a luminescent DEVD peptide substrate for caspase-3/7 and a recombinant thermostable luciferase is added to detect apoptosis.
